Rapid in viro multiplication of some sugarcane cultivars (saccharum officnarum) via embryogenic callus culture of young leaf tissues

Abstract: 

In this study, we have reported an efficient protocol for induction of callus and regeneration without sterliztion step of some sugarcane cultivars. Callus induction from young leaf tissues was attained from MS culture medium supplemented with different concentrations of the growth regulator substances. The results showed that the highest callus rate on MS medium supplemented with 3-4mg/l 2,4D. The highest multiplication of arpical meristem was observed on MS medium which was supplemented with BAP 2-2.5 mg/l, kinetin 0,2 mg/l, 10mg/l PVP and 15% coconut juice. Medium culture supplemented with BAP 0,5mg/l for pre-root prolongation was conformed for shoot growth with height of 5-6 cm in uniform. The most suitable concentration of NAA was 2.5mg/l in liquid culture which enhanced rhizogenesis over 90%. The optimal subtrates for releasing plant were included 1 alluvial soil +1 clean humus + ¼ microorganic fertilizer + 1 sand were shown the best survival rate of plants over 90%. This in vitro regeneration techniques was prompt and cost-effective and possibly used for large scale in vitro regeneration of sugarcane cultivars.
